Target
This gene encodes an ion channel that is activated by high temperatures above 52 degrees Celsius. The protein may be involved in transduction of high-temperature heat responses in sensory ganglia. It is thought that in other tissues the channel may be activated by stimuli other than heat.
FUNCTION: Calcium-permeable, non-selective cation channel with an outward rectification. Seems to be regulated, at least in part, by IGF-I, PDGF and neuropeptide head activator. May transduce physical stimuli in mast cells. Activated by temperatures higher than 52 degrees Celsius; is not activated by vanilloids and acidic pH.
SUBCELLULAR LOCATION: Cell membrane; Multi-pass membrane protein. Cytoplasm. Melanosome. Note: Translocates from the cytoplasm to the plasma membrane upon ligand stimulation. Identified by mass spectrometry in melanosome fractions from stage I to stage IV.

Storage
Maintain the lyophilised/reconstituted antibodies frozen at -20°C for long term storage and refrigerated at 2-8°C for a shorter term. When reconstituting, glycerol (1:1) may be added for an additional stability. Avoid freeze and thaw cycles.

IHC on paraffin sections of human large intestine tissue using Rabbit antibody to TRPV2 (550-600): OST00182W. HIER: 1 mM EDTA, pH 8 for 20 min using Thermo PT Module. Blocking: 0.2% LFDM in TBST filtered thru 0.2 µm. Detection was done using Novolink HRP polymer from Leica following manufacturer's instructions. Primary antibody: dilution 1: 1000, incubated 30 min at RT (using Autostainer). Sections were counterstained with Harris Hematoxylin. |
