Immunogen
A synthetic peptide from the internal region of human HOMER1 (Home1, Homer protein homolog 1, SYN47) conjugated to an immunogenic carrier protein was used as the antigen. The peptide is homologous with the corresponding sequence in mouse, rat and monkey.

Target
FUNCTION: Postsynaptic density scaffolding protein. Binds and cross-links cytoplasmic regions of GRM1, GRM5, ITPR1, DNM3, RYR1, RYR2, SHANK1 and SHANK3. By physically linking GRM1 and GRM5 with ER-associated ITPR1 receptors, it aids the coupling of surface receptors to intracellular calcium release. May also couple GRM1 to PI3 kinase through its interaction with CENTG1. Isoform 1 regulates the trafficking and surface expression of GRM5. Isoform 3 acts as a natural dominant negative, in dynamic competition with constitutively expressed isoform 1 to regulate synaptic metabotropic glutamate function. Isoform 3, may be involved in the structural changes that occur at synapses during long-lasting neuronal plasticity and development.

Storage
Maintain the lyophilised/reconstituted antibodies frozen at -20°C for long term storage and refrigerated at 2-8°C for a shorter term. When reconstituting, glycerol (1:1) may be added for an additional stability. Avoid freeze and thaw cycles.

IHC-P on paraffin sections of rat spinal cord. The animal was perfused using Autoperfuser at a pressure of 110 mmHg with 300 ml 4% FA and further post fixed overnight before being processed for paraffin embedding. HIER: Tris-EDTA, pH 9 for 20 min using Thermo PT Module. Blocking: 0.2% LFDM in TBST filtered thru 0.2 µm. Detection was done using Novolink HRP polymer from Leica following manufacturers instructions. Primary antibody: dilution 1: 1000, incubated 30 min at RT using Autostainer. Sections were counterstained with Harris Hematoxylin. |
